About JEMPERLI® Infusion Therapy

JEMPERLI® (dostarlimab-gxly) is a biologic medication known as a PD-1 blocking antibody, given as an intravenous (IV) infusion. Rather than attacking cancer cells directly, it blocks a signal that tumors use to hide from your immune system, which helps your own immune cells recognize and attack the cancer.

JEMPERLI® is typically given as a 30-minute infusion: 500 mg every three weeks for the first six cycles, then 1,000 mg every six weeks after that. Depending on your diagnosis, the early cycles may be given alongside carboplatin and paclitaxel chemotherapy. Your oncologist sets the schedule.

Conditions treated by this therapy

JEMPERLI® is FDA-approved for the treatment of certain adult cancers, used either alone or in combination with chemotherapy. Your care team may consider this therapy if you have:

Primary advanced or recurrent endometrial cancer, given with carboplatin and paclitaxel and then continued on its own
Mismatch repair deficient (dMMR) recurrent or advanced endometrial cancer that has progressed after a platinum-containing regimen, when curative surgery or radiation is not an option
Mismatch repair deficient (dMMR) recurrent or advanced solid tumors that have progressed after prior treatment, when there is no satisfactory alternative option

Eligibility for the dMMR indications is confirmed with an FDA-approved test. Your oncologist will confirm whether JEMPERLI® is right for you, and which regimen fits your diagnosis, before treatment begins.

What to Expect During JEMPERLI® Infusion Therapy

When you arrive at our Violet Health location, you'll settle into a private, welcoming space designed with your comfort and peace of mind in mind. Before and during your intravenous infusion, our experienced team will:

Review your current health status and any symptoms you've noticed
Check your vital signs and review recent lab work, including thyroid, liver, and kidney function, before treatment begins
Place your IV using gentle, practiced technique
Administer your JEMPERLI® infusion over about 30 minutes with our nursing team at your side
Monitor you closely for any infusion-related reactions
Protect your privacy and comfort throughout the visit

JEMPERLI® is given every three weeks for the first six cycles, then every six weeks, which means fewer infusion visits as treatment continues. When JEMPERLI® is given on the same day as carboplatin and paclitaxel, it is administered first, so plan to be with us for a few hours. Visits where JEMPERLI® is given on its own are considerably shorter.

JEMPERLI® Side Effects

As with any medication, JEMPERLI® may cause side effects. Being informed helps you know what to watch for and when to contact your healthcare team. Common Side Effects:

Fatigue or weakness
Low red blood cell count (anemia)
Nausea and vomiting
Diarrhea or constipation
Rash
Numbness or tingling in the hands and feet (peripheral neuropathy), when given with chemotherapy
Hair loss, when given with chemotherapy
Joint pain
Decreased appetite and abdominal pain
Shortness of breath
Urinary tract infection
Changes in blood counts, liver enzymes, kidney function, blood sugar, and electrolyte levels

JEMPERLI® can cause your immune system to attack healthy organs and tissues. These immune-mediated reactions can become severe or life-threatening, can affect any organ system, and may occur during treatment or after it has ended. Please alert our clinical staff or seek medical attention immediately if you experience any of the following:

Lung inflammation (pneumonitis), including new or worsening cough, chest pain, or shortness of breath
Intestinal inflammation (colitis), including diarrhea, stool changes, severe abdominal pain, or blood or mucus in the stool
Liver problems (hepatitis), including yellowing of the skin or eyes, dark urine, severe nausea, or pain on the right side of the abdomen
Hormone gland problems affecting the thyroid, adrenal glands, or pituitary gland, including headaches, extreme tiredness, weight changes, dizziness or fainting, constipation, hair loss, or feeling cold
New or worsening diabetes, including increased thirst or hunger, frequent urination, and unexplained weight loss
Kidney problems (nephritis), including changes in the amount or color of your urine or swelling in the ankles
Serious skin reactions, including rash, blistering or peeling skin, painful sores or ulcers in the mouth, nose, throat, or genital area, fever or flu-like symptoms, and swollen lymph nodes
Infusion reactions, including chills or shaking, itching or rash, flushing, wheezing, dizziness, feeling faint, fever, and back or neck pain
Other immune-mediated problems affecting the heart, nerves, eyes, muscles, or pancreas, including irregular or rapid heartbeat, muscle pain or weakness, eye problems or vision changes, and numbness and tingling

Stem cell transplant: Tell your care team if you have had, or are planning to have, an allogeneic stem cell transplant. Serious complications can occur in patients treated with a PD-1 blocking antibody before or after transplant.

Pregnancy warning: JEMPERLI® may harm an unborn baby. Tell your care team if you are pregnant or plan to become pregnant. Use effective birth control during treatment and for four months after your last dose, and do not breastfeed during treatment or for four months after your last dose.
